G-418 disulphate (Geneticin) is an aminoglycoside antibiotic widely used as a selective agent in eukaryotic cell culture. It exhibits activity against both prokaryotic and eukaryotic cells by inhibiting protein synthesis. G-418 is most commonly used for the selection and maintenance of genetically engineered cells expressing the neomycin resistance gene (neo), making it a key reagent in stable cell line development and genetic engineering workflows. Mechanism of Action: G-418 binds to the 80S ribosome in eukaryotic cells and the 70S ribosome in prokaryotic cells, causing misreading of mRNA and inhibition of protein synthesis. This leads to the production of nonfunctional proteins and ultimately cell death.
