Methotrexate (MTX) is a folate analog and potent antimetabolite widely used in molecular and cell biology research. It inhibits dihydrofolate reductase (DHFR), a key enzyme in nucleotide biosynthesis, and is commonly used to study cell proliferation, DNA synthesis, and apoptosis. Methotrexate is also a critical reagent in DHFR-based gene amplification systems, particularly in CHO cell expression platforms for recombinant protein production. Mechanism of Action: Methotrexate inhibits dihydrofolate reductase (DHFR), preventing the conversion of dihydrofolate to tetrahydrofolate, which is required for thymidylate and purine synthesis. This leads to inhibition of DNA replication and cell division. In DHFR-based systems, methotrexate applies selective pressure that drives amplification of the DHFR gene and linked transgenes, enabling high-level protein expression.
