Trimethoprim lactate is a synthetic diaminopyrimidine antibiotic widely used in microbiology and molecular biology research. It exhibits antibacterial activity by selectively inhibiting bacterial dihydrofolate reductase (DHFR), a key enzyme in folate metabolism. Trimethoprim is commonly used alone or in combination with sulfonamides such as sulphamethoxazole to achieve synergistic inhibition of bacterial growth. Mechanism of Action: Trimethoprim selectively inhibits bacterial dihydrofolate reductase (DHFR), preventing the reduction of dihydrofolic acid to tetrahydrofolic acid. This disrupts the synthesis of nucleotides and ultimately inhibits DNA, RNA, and protein synthesis. When combined with sulfonamides, trimethoprim provides a sequential blockade of folate biosynthesis, resulting in enhanced antibacterial efficacy.
