Mercaptopurine monohydrate (6-mercaptopurine, 6-MP) is a thiopurine analog widely used as an antimetabolite in molecular biology and cell biology research. It interferes with purine nucleotide metabolism and is commonly used to study DNA synthesis, cell proliferation, and apoptosis. Mercaptopurine is also used in genetic selection systems, particularly in combination with hypoxanthine-guanine phosphoribosyltransferase (HGPRT)-based selection. Mechanism of Action: Mercaptopurine is converted intracellularly into active thioguanine nucleotides, which inhibit de novo purine synthesis and become incorporated into DNA and RNA. This disrupts nucleic acid synthesis and function, ultimately inhibiting cell proliferation and inducing cell death.
