Mitomycin C is a potent antitumor antibiotic isolated from Streptomyces caespitosus, widely used in molecular and cell biology research as a DNA crosslinking agent. It is commonly applied to study DNA damage response, cell cycle regulation, and apoptosis. Mitomycin C is also frequently used to inhibit cell proliferation in co-culture systems, such as feeder cell preparation. Mechanism of Action: Mitomycin C is a bioreductive alkylating agent that becomes activated intracellularly and forms covalent crosslinks between DNA strands. These crosslinks inhibit DNA replication and transcription, ultimately leading to cell cycle arrest and apoptosis.
