Bleomycin sulphate is a glycopeptide antibiotic with potent antitumor and DNA-cleaving activity, originally isolated from Streptomyces verticillus. It is widely used in molecular and cell biology research as a tool to induce DNA strand breaks and study DNA damage response pathways. Bleomycin is also used as a selective agent in eukaryotic systems through resistance genes such as ble, which confer protection against its cytotoxic effects. Mechanism of Action: Bleomycin binds to DNA and, in the presence of oxygen and metal ions, induces single- and double-strand DNA breaks through free radical formation. This results in inhibition of DNA synthesis and triggers apoptosis or cell cycle arrest. Resistance is conferred by bleomycin-binding proteins encoded by genes such as ble, which inactivate the drug by preventing its interaction with DNA.
