Acyclovir is a synthetic purine nucleoside analog with potent antiviral activity, particularly against herpes simplex virus (HSV) and varicella-zoster virus (VZV). It is widely used in virology and cell biology research to inhibit viral replication and to study virus–host interactions. Acyclovir is also employed as a selective agent in systems utilizing herpes simplex virus thymidine kinase (HSV-TK) as a negative selection marker. Mechanism of Action: Acyclovir is selectively phosphorylated by viral thymidine kinase (TK) to its monophosphate form, and subsequently converted to the active triphosphate by cellular kinases. Acyclovir triphosphate inhibits viral DNA polymerase and is incorporated into viral DNA, leading to chain termination and inhibition of viral replication.
