Doxycycline hydrochloride is a broad-spectrum tetracycline antibiotic widely used in microbiology, molecular biology, and cell biology applications. It exhibits activity against a wide range of Gram-positive and Gram-negative bacteria and is best known as a key inducer in tetracycline-regulated gene expression systems (Tet-on/Tet-off). Due to its high stability and cell permeability, doxycycline is the preferred tetracycline derivative for inducible gene expression studies. Mechanism of Action: Doxycycline inhibits bacterial protein synthesis by binding to the 30S ribosomal subunit, preventing the attachment of aminoacyl-tRNA to the ribosome and blocking translation. In inducible gene expression systems, doxycycline binds to tetracycline-controlled transactivators (rtTA/tTA), regulating transcription of target genes in a dose-dependent manner.
