Chloramphenicol is a broad-spectrum antibiotic belonging to the amphenicol class, originally isolated from Streptomyces venezuelae. It exhibits activity against a wide range of Gram-positive and Gram-negative bacteria and is widely used in molecular biology and microbiology applications. Chloramphenicol is commonly employed as a selective agent in cloning and protein expression systems, particularly in plasmids carrying the cat gene. Mechanism of Action: Chloramphenicol inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit, blocking peptidyl transferase activity and preventing peptide bond formation. This results in inhibition of protein elongation and ultimately suppresses bacterial growth. Resistance is typically mediated by chloramphenicol acetyltransferase (CAT), which inactivates the antibiotic, as well as efflux pumps or reduced membrane permeability.
