Linsmaier & Skoog (LS) Medium was developed in 1965 to optimize the organic requirements of tobacco (Nicotiana) tissue cultures. The formulation is based on Murashige & Skoog (MS) basal salts supplemented with a modified vitamin composition defined by Linsmaier and Skoog. It provides a balanced nutrient system of inorganic salts and vitamins to support plant cell growth and differentiation. Nitrogen is supplied as potassium nitrate and ammonium nitrate, contributing to cell proliferation and pH stability. The LS vitamin formulation includes an increased thiamine concentration (0.4 mg/L), compensating for a simplified vitamin system and supporting metabolic activity. LS Medium is widely used across plant tissue culture applications.
