Kao & Michayluk Medium, as described in 1975, was originally developed for the culture of protoplasts of Vicia hajastana. The formulation contains the macro- and micronutrients defined by Kao and Michayluk and is optimized to support protoplast viability, cell wall regeneration, and subsequent cell division. When the mineral salt medium was enriched with organic acids, sugars, sugar alcohols, growth regulators, amino acids and other organic compounds. It is particularly effective for cultures initiated at very low cell densities (1–2 cells/mL). Increasing calcium chloride concentration from 1 mM to 5 mM has been shown to further stimulate cell growth and division.
