Gresshoff & Doy Medium, as described in 1974, was developed for the growth of haploid callus and plantlets derived from anthers of Arabidopsis thaliana. The system was optimized for anthers excised at late prophase of meiosis, a developmental stage favorable for haploid induction. The defined mineral formulation supports callus formation under high auxin–low kinetin conditions. Subsequent differentiation is achieved by transferring tissues to a low auxin–high kinetin medium under a light–dark cycle. This medium has also been successfully applied to haploid callus cultures of tomato, barley, and Vitis vinifera.
