Gamborg B5 Medium was developed by Gamborg et al. (1968) for the initiation and growth of soybean (Glycine max) cell suspension cultures. The formulation provides defined macro- and micronutrients optimized for active plant cell proliferation. Unlike MS medium, B5 contains no ammonium nitrate; instead, nitrogen is supplied as ammonium sulfate and elevated levels of potassium nitrate. Ammonium concentrations above 2 mM were shown to inhibit cell growth, and the B5 formulation reflects this optimization. B5 medium has become a widely used nutrient system for plant cell lines and tissue cultures.
